Abstract

[Objective]The community composition and diversity characteristics of endophytic bacteria in Rhizoma Musae were explored and the correlations between endophytic bacteria and active ingredients in Rhizoma Musae were analyzed to provide the scientific basis for improving quality of Rhizoma Musae.[Method]The endophytic bacterial communities in Rhizoma Musae and metabolic products of the medicinal materials were analyzed by 16S rDNA high-throughput sequencing and liquid chromatography-mass spectrometry(LC-MS)technique.The endophytic bacterial groups closely related with metabolic products and active ingredients of Rhizoma Musae were screened by Pearson correlation analysis.[Result]At phylum level,the relative abundance of Mycoplasmatota,Pseudomonadota and Actinomycetota was 63.42%-86.59%,7.84%-31.52%and 1.02%-2.63%respectively.At genus level,Candidatus Phytoplasma was a dominant species and its relative abundance reached 63.42%-86.59%.117 mutual differential metabolites(DEMs)including glycoside,glycolipids,polyketone,terpene,sterides and so on were screened from Rhizoma Musae samples collected from 5 Rhizoma Musae producing areas and mainly enriched in metabolic pathways of ABC transport protein,flavonoid biosynthesis,amino acid biosynthesis.The correlation analysis showed that Inquilinus was significantly and positively related to total phenolic components,and significantly and negatively related to flavonoids compounds.Catenulispora and Mycobacterium were significantly and positively related to total alkaloids components.[Conclusion]The community structure and diversity of endophytic bacteria in Rhizoma Musae from different producing areas are relatively stable.There are differences in the relative abundance of the dominant endophytic bacteria.There are significant correlations between bacterial genera of endophytic bacteria and metabolites of Rhizoma Musae.The specific bacterial genera may involve in synthesis or conversion process of active ingredients by means of regulating key metabolic pathway.The synthesis of secondary metabolites such as flavone and alkaloid in Rhizoma Musae may be closely related to endophytic bacteria.
