Evaluation of Polysaccharide Quality Between Magoderna subresinosum(Murrill)Steyaert and Six Species of Ganoderma Medicinal Based on Characteristic Chromatograms and Content Determination

Abstract
Objective To establish characteristic chromatograms of monosaccharides derived from Magoderna subresinosum(Murrill)Steyaert and determine the contents of its main monosaccharides,thereby exploring the differences between M.subresinosum(Murrill)Steyaert and six other Ganoderma species.Methods Crude polysaccharides were extracted using water extraction and alcohol precipitation.The hydrochloric acid hydrolysates were derivatized with 1-phenyl-3-methyl-5-pyrazolone(PMP)and analyzed by high-performance liquid chromatography(HPLC)to generate characteristic chromatograms.The contents of D-mannose,D-glucose,and galactose were determined using the internal standard method.Results Six characteristic peaks were identified and annotated across 23 batches of M.subresinosum(Murrill)Steyaert and 24 batches of six other Ganoderma species[Ganoderma lucidum(Leyss.ex Fr.)Karst,G.sinense Zhao,Xu et Zhang,G.daipingshanense J.D.Zhao,G.applanatum(Pers.)Pat,G.applanatum(Pers.ex Gray)Pat,G.atrum].These peaks corresponded to D-mannose,D-glucuronic acid,D-glucose,galactose,D-xylose,and L-fucose.D-glucose exhibited the highest peak intensity,followed by D-mannose and galactose,while D-xylose showed the weakest intensity.However,the peak area ratio of galactose to D-glucose demonstrated notable variation.The peak area ratio for M.subresinosum(Murrill)Steyaert((x)±s)was 0.126±0.046,which was generally lower than that of G.sinense Zhao,Xu et Zhang and G.atrum but higher than that of G.lucidum(Leyss.ex Fr.),G.daipingshanense J.D.Zhao,G.applanatum(Pers.)Pat,G.applanatum(Pers.ex Gray)Pat.The mean contents((x)±s)of D-mannose,D-glucose,and galactose,along with their total content in G.capense,were 0.134%±0.059%,0.972%±0.270%,0.113%±0.038%,and 1.225%±0.318%,respectively.The average total monosaccharide content in descending order was G.applanatum(Pers.ex Gray)Pat,Ganoderma capense(Lioyd)Teng,M.subresinosum(Murrill)Steyaert,G.sinense Zhao,Xu et Zhang,G.atrum,G.lucidum(Leyss.ex Fr.)Karst,G.daipingshanense J.D.Zhao,G.applanatum(Pers.)Pat.Conclusion The pre-column derivatization HPLC method for determining D-mannose,D-glucose,and galactose is stable and feasible,and can be effectively used to evaluate the quality differences in polysaccharides between M.subresinosum(Murrill)Steyaert and six other Ganoderma species.关键词
