Abstract

To investigate the differences in disease resistance between Larimichthys crocea and Nibea albi-flora against Pseudomonas plecoglossicida infection,as well as the immune response dynamics of their skin dur-ing the infection process,both immersion and intraperitoneal injection methods were used to challenge the fish.Skin tissues were collected at 3,24,and 48 h after immersion challenge for transcriptome sequencing analysis.The results showed that N.albiflora exhibited significantly lower morbidity and mortality than L.crocea,indica-ting a stronger resistance to infection.Following immersion challenge,L.crocea began to show disease symptoms and mortality by the third day,with a morbidity rate of 65.3%and mortality rate of 37.5%by the seventh day,respectively,whereas N.albiflora showed no signs of disease or death.After injection challenge,the mor-bidity and mortality rates of L.crocea reached 96.5%and 68.3%by the seventh day,respectively,while those of N.albiflora were significantly lower at 55.5%and 38.3%,respectively.In L.crocea,a strong immune acti-vation was observed as early as 3 h post-infection,with a large number of differentially expressed genes en-riched in inflammatory response pathways such as cytokine-cytokine receptor interaction,IL-17 signaling,and NF-κB signaling,and this high level of immune activation was maintained throughout the infection period.In contrast,N.albiflora exhibited a milder and more regulated immune response,with lower activation of immune-related pathways and a transition toward neuro-regulation and metabolic homeostasis pathways after 24 h.
