水稻NRT1/PTR基因的全基因组鉴定及其响应酸胁迫的表达分析
Genome-wide identification of rice NRT1/PTR genes and their expression analysis in response to acid stress

Abstract
[Objective]The study aimed to conduct a genome-wide identification of the nitrate transporter 1/peptide transporter(NRT1/PTR)family genes in rice,elucidate their response mechanisms to acid stress,providing theoretical basis for molecular improvement of nitrogen utili-zation efficiency in rice.[Method]Bioinformatic approaches were employed to identify NRT1/PTR family genes at the genome-wide level in rice,and to analyze their protein physicochemical properties and gene duplication events.Based on transcriptome data,the tissue-specific ex-pression patterns of OsNRT1/PTR genes and their expression patterns under nitrogen treatments and acid stresses were analyzed.Key candi-date genes whose expression patterns in roots under acid stresses and nitrogen treatments remained highly consistent with those under normal conditions were screened and subsequently validated by real-time fluorescence quantitative PCR.[Result]A total of 90 OsNRT1/PTR genes identified at the genome-wide level in rice,whose expansion mainly came from segmental and tandem duplication events and underwent strong purifying selection.Transcriptome data analysis demonstrated that OsNRT1/PTR genes exhibited obvious tissue-specific expression patterns and diverse stress responses;genes showed significant up-regulation or down-regulation under nitrogen treatments or acid stresses(P<0.05),with some genes exhibiting genotype-specific response patterns.Ten key candidate genes(LOC_Os01 g68510,LOC_Os10g42870,LOC_Os10g22560,LOC_Os02g52060,LOC_Os06g15370,LOC_Os01g65100,LOC_Os01g37590,LOC_Os08g05910,LOC_Os01g65110,LOC_Os04g50950)maintaining consistent response patterns under acid stress and nitrogen conditions with those in roots under normal conditions were screened through co-expression analysis.Real-time fluorescence quantitative PCR analysis revealed marked differences in the responses to acid stresses and nitrogen treatments of OsNRT1/PTR genes from different rice varieties.[Conclusion]O.sNRT1/PTR genes exhibit diverse and complex regulatory patterns in response to nitrogen treatments and acid stresses.The 10 key candidate genes identified may play central roles in maintaining functional stability in roots and enhancing plant adaptability to stress.关键词
