lncRNA ADIRF-AS1 promotes intrauterine adhesions by regulating epithelial-mesenchymal transition in human endometrial epithelial cells through the miR-185-5p/SIX4 axis

Abstract
Objective To investigate the effect of the long non-coding RNA(lncRNA)ADIRF-AS1 on the proliferation of human endo-metrial epithelial cells(HEECs)and its underlying mechanism in intrauterine adhesions(IUA).Methods Endometrial tissues and peripheral blood samples were collected from 20 patients with moderate-to-severe IUA and 20 healthy individuals with normal endome-trium(control group)who visited the Department of Reproductive Medicine in our hospital between January 2022 and June 2024.Quan-titative real-time PCR(qRT-PCR)was performed to determine ADIRF-AS1 expression levels in endometrial tissues,peripheral blood,and HEECs treated with transforming growth factor-β1(TGF-β1).Bioinformatics analysis and dual-luciferase reporter assays were used to verify the relationship between the ADIRF-AS1 and miR-185-5p.qRT-PCR and Western blotting were performed to measure the expres-sion of epithelial-mesenchymal transition(EMT)-related proteins in HEECs transfected with miR-185-5p or SIX4.Results Compared with the control group,ADIRF-AS1 expression was significantly upregulated in endometrial tissues and peripheral blood of patients with moderate-to-severe IUA and TGF-β1-stimulated human endometrial cells(P<0.05).High ADIRF-AS1 expression promoted the pro-liferation,invasion,and migration of HEECs(P<0.05).Notably,the ADIRF-AS1 sponged miR-185-5p.Downregulation of miR-185-5p promoted the proliferation,invasion,and migration of HEECs by targeting SIX4 to mediate EMT,thereby facilitating the occurrence and progression of IUA.Conclusion The lncRNA ADIRF-AS1 promoted the proliferation of HEECs by regulating EMT through the miR-185-5p/SIX4 axis,thus contributing to the development of IUA.关键词
