Abstract
[Objective]To investigate the expression levels of extracellular matrix protein 1(ECM1)and four-jointed box kinase 1(FJX1)in colorectal cancer(CRC)tissues,and to analyze their predictive value for 5-year prognosis after surgery.[Methods]A total of 92 CRC patients who were diagnosed and treated in Puyang Oilfield General Hospital from January 2018 to January 2020 were prospectively selected as the research objects.The mRNA and protein expression levels of ECM1 and FJX1 in cancer tissues and adjacent non-cancerous tissues were compared.The expression levels of ECM1 and FJX1 proteins in cancer tissues of patients with different clinical characteristics were analyzed.According to the prognosis of 5 years after operation,CRC patients were divided into poor prognosis subgroup and good prognosis subgroup,and the mRNA and protein expression levels of ECM1 and FJX1 in cancer tissues were compared.The interaction effect of ECM1 and FJX1 in the 5-year prognosis of CRC patients was analyzed.The predictive value of ECM1 and FJX1 for 5-year prognosis after surgery was evaluated.[Results]The mRNA and protein expression levels of ECM1 and FJX1 in cancer tissues were higher than those in adjacent tissues(P<0.05).The mRNA and protein expression levels of ECM1 and FJX1 in the poor prognosis subgroup were higher than those in the good prognosis subgroup(P<0.05).According to the average level of ECM1 and FJX1 protein expression in patients,they were divided into high level and low level,and four subgroups were obtained,which were low level of ECM1+low level of FJX1(R00),low level of ECM1+high level of FJX1(R01),high level of ECM1+low level of FJX1(R10),high level of ECM1+high level of FJX1(R11).R00 group was used as reference,R01,R10 and R11 could increase the risk of poor prognosis in 5 years after operation by 2.014,3.290 and 6.327 times(P<0.05).The expression levels of ECM1 and FJX1 proteins had a positive additive interaction effect on 5-year postoperative poor prognosis,and AP=0.3197.The proportion of postoperative poor prognosis caused by the interaction between the two was 31.97％.The area under the receiver operating characteristic curve(AUC)value of combined prediction of ECM1 and FJX1 protein expression levels for 5-year postoperative prognosis was higher than that of single index prediction(P<0.05).[Conclusion]The expression levels of ECM1 and FJX1 in CRC cancer tissues are significantly higher than those in adjacent non-cancerous tissues.In patients with poor 5-year postoperative prognosis,the expression levels of ECM1 and FJX1 are notably elevated than those with good prognosis.The expression levels of ECM1 and FJX1 proteins exhibit a positive additive interaction effect on postoperative poor prognosis.Measuring their levels holds predictive value for poor 5-year postoperative prognosis,and combining their levels for detection offers even higher predictive value.关键词
