摘要

Abstract

Sensorineural hearing loss is the most prevalent form of hearing impairment,and its therapeutic outcomes are largely limited by inefficient drug delivery to the inner ear.Systemic administration,including oral and intravenous routes,leads to poor drug penetration into the inner ear owing to the presence of the blood-labyrinth barrier.Although local delivery approaches,such as intratympanic or round window administration,can increase cochlear drug concentrations,they often produce a base-to-apex concentration gradient,which hampers precise intervention at specific cochlear regions and target cells.Magnetically controlled inner ear drug delivery employs externally applied magnetic fields to guide drug-loaded magnetic nanoparticles across anatomical barriers and enable their directed transport within the inner ear.This strategy may help mitigate uneven intracochlear drug distribution and provides a potential approach for more precise treatment of inner ear diseases.
