Abstract

Objective To construct a mitochondria-targeted near-infrared aggregation-induced emission(AIE)probe,TSPH,and to evaluate its performance for the real-time dynamic imaging of viscosity in living cells.Methods Using triphenylamine as the core structure,a thiophene ring and a pyridinium salt targeting unit were successively introduced via Suzuki coupling and Knoevenagel condensation reactions to synthesize the probe TSPH,which was characterized by nuclear magnetic resonance(NMR)spectroscopy and high-resolution mass spectrometry(HRMS).Its AIE properties,viscosity response,selectivity,and photostability/pH stability were systematically studied by spectroscopic methods.Cytotoxicity was assessed using the CCK-8 assay,mitochondrial targeting was verified by co-localization experiments,and confocal imaging was employed to monitor the probe's response to nystatin-induced changes in mitochondrial viscosity.Results TSPH exhibited typical AIE characteris-tics.Within the viscosity range of 0.59 to 945 cP,its fluorescence intensity showed a good linear relationship with the logarithm of viscosity(Log F=0.364 1 Log η+1.751 1,R2=0.987 2).The probe demonstrated ex-cellent photostability and was unaffected by pH variations(4.0-9.0)or common biomolecules.Cellular experi-ments indicated low cytotoxicity of TSPH,with a co-localization coefficient of 0.83 compared to a commercial mi-tochondrial dye,confirming specific mitochondrial targeting.The probe could sensitively and dynamically monitor nystatin-induced changes in mitochondrial viscosity,with fluorescence intensity positively correlated with drug concentration and treatment time.Conclusion A mitochondria-targeted near-infrared AIE probe,TSPH,was successfully constructed,providing an effective imaging tool for studying organelle function and exploring the mechanisms of viscosity-related diseases.
