不同基原地骨皮HPLC指纹图谱与4种成分的含量测定研究
Study on HPLC Fingerprints and Quantitation Analysis of Four Components in Different Botanical Origins of Lycii Cortex

Abstract
Objective:To analyze the differences in chemical components between two types of Lycii cortex derived from Lycium chinense Mill.and Lycium barbarum L.Methods:HPLC was used to establish the fingerprint profiles of the two kinds of Lycii cortex samples,and the common pattern diagrams were compared for similarity analysis.An HPLC method was developed for the determination of four components:(E)-N-caffeoylputrescine,scopoline,kukoamine B,kukoamine A.Combined with principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA),the differences in chemical components of Lycii cortex from different origins were compared.Results:HPLC fingerprints of Lycii cortex from different origins were established.Lycii cortex originated from Lycium chinense Mill.had 14 common peaks,while Lycii cortex originated from Lycium barbarum L.had 11 common peaks.Five common peaks were identified:(E)-N-caffeoylputrescine(Peak 1),scopolin(Peak 2),kukoamine B(Peak 3),kukoamine A(Peak 5),and scopoletin(Peak 6).The quantitative analysis showed good linearity for the four components within the concentration ranges of 1.37~87.80 μg·mL-1(r=0.999 9),1.97~125.83 μg·mL-1(r=0.999 7),6.31~403.76 μg·mL-1(r=0.999 9),and 10.67~341.57 μg·mL-1(r=0.999 8),with average recoveries of 100.52%~103.84%and RSD of 2.33%~3.42%.The content determination results showed that the average percentages of the four components in Lycii cortex originated from Lycium barbarum L.were higher than in Lycium chinense Mill.Chemical pattern recognition analysis classified the two types of Lycii cortex into two categories,and(E)-N-caffeoylputrescine played a prominent role in model construction.Conclusion:The established method is accurate and reliable,which can be utilized to distinguish the two varieties of Lycii Cortex and provide a reference for the quality control of them.关键词
