李属坏死环斑病毒甜樱桃分离物的全基因组克隆及遗传变异分析
Complete Genome Sequences and Phylogenetic Analysis of a Sweet Cherry Isolate of Prunus necrotic Ringspot Virus

Abstract
Taking sweet cherry leaf samples exhibiting typical viral disease symptoms with necrotic spots,enations,and leaf curling as the test materials,RT-PCR combined with 5'/3'rapid amplification of cDNA ends(RACE)technology was used to obtain the complete genome sequence of a Prunus necrotic ringspot virus(PNRSV)isolate from Yantai.The genetic variation of the PNRSV genome was studied,in order to provide references for the development of efficient molecular detection techniques for PNRSV,understanding the epidemiology of PNRSV in major sweet cherry production areas in China,and formulating targeted control strategies.The results showed that the genome of the isolate consists of three positive-sense single-stranded RNAs,RNA1 was 3 338 nt in length,RNA2 was 2 581 nt,and RNA3 was 1 949 nt.The genome structure was highly consistent with known PNRSV isolates.Phylogenetic analysis based on the coat protein(CP)gene and movement protein(MP)gene revealed that the PNRSV population exhibits significant genetic diversity worldwide.Isolates from different geographical origins form multiple branches in the phylogenetic tree,demonstrating a complex pattern of geographical distribution and population differentiation.关键词
