摘要

Abstract

To explore the antibacterial activity of carbon nanotube(CNT)nanomaterials adsorbed with flor-fenicol(FF)against intracellular Salmonella typhimurium(ST)infection.Using mouse mononuclear mac-rophage(RAW264.7)as the model cell,a model of intracellular infection of RAW264.7 cells by Salmonel-la typhimurium was constructed.And the conditions such as the multiplicity of infection(MOI),the clear-ance of extracellular bacteria,and the lysis conditions were respectively optimized;In this experiment,the minimum inhibitory concentration values of FF and CNT-FF complexes against ST and the cytotoxicity test were determined to evaluate their anti-intracellular ST effects and biocompatibility.The results showed that the intracellular infection system of Salmonella typhimurium was composed of multiple infections with MOI=200 infecting cells for 1 h.Gentamicin at 50 μg/mL was administered for 0.5 h to kill the extra-cellular bacteria,and the lysis buffer was PBS containing 0.02%Tween-20.The intracellular bacterial clearance test indicated that the clearance rate of intracellular ST in the CNT-modified FF group(66%)was significantly higher than that in the FF group(24%)(P<0.05);The cytotoxicity tests indicated that when the concentration of the CNT-FF complex was not higher than 30 μg/mL,it had no effect on cell sur-vival rate,showing good biocompatibility.In this study,a model of intracellular infection of mononuclear macrophages by Salmonella typhimurium was successfully constructed.The CNT-FF complex showed strong antibacterial activity against intracellular ST,which can provide a reference basis for formulating a reasonable drug administration plan for the treatment of intracellular ST.
