Construction of A 3D Breast Cancer Spheroid Model and Synchrotron X-ray Fluorescence Imaging Analysis of Its Spatial Elemental Distribution
摘要
Abstract
Compared with conventional two-dimensional(2D)adherent cell culture,three-dimensional(3D)tumor spheroids can better recapitulate the spatial architecture and microenvironmental characteristics of tumor tissues, thus providing a more physiologically relevant model for investigating elemental metabolism in cancer cells. In this study,a 3D breast cancer spheroid culture system was established using agarose-coated U-bottom plates, and anin situ analytical method was developed for characterizing the spatial distribution of multiple elements within tumor spheroids by combining this model with synchrotron micro-X-ray fluorescence(micro-XRF)imaging.The results showed that among the three types of tested breast cancer cell lines, namely 4T1,MCF-7,and MDA-MB-231,MDA-MB-231 cells exhibited the best spheroid-forming ability and generated compact spheroids with clear boundaries and controllable sizes.Synchrotron micro-XRF imaging analysis revealed that, compared with 2D-cultured cells, the signals of Ca,Fe,and Zn were significantly enhanced in 3D tumor spheroids and displayed a spatially heterogeneous distribution pattern characterized by peripheral enrichment and relative depletion in the central region. This study established an analytical platform integrating a 3D tumor spheroid model with synchrotron micro-XRF elemental imaging, providing an effective approach for investigating tumor-associated metal element metabolism under conditions that more closely resemble the in vivo microenvironment.关键词
