Effects of propofol on spontaneous calcium oscillations in human monocyte-derived microglia-like cells

Abstract
Objective To establish a human monocyte-derived microglia-like cell(MDMi)model,character-ize its spontaneous calcium oscillatory activity,and investigate the effects of the anesthetic agent propofol on calcium signa-ling in MDMi.Methods Peripheral blood monocytes were isolated and differentiated into MDMi over 14 days using cyto-kine induction.Cellular identity was validated through morphological assessment,immunofluorescence staining,and in-flammatory cytokine release following lipopolysaccharide(LPS)stimulation.Calcium imaging was employed to record spontaneous calcium transients,and the effects of propofol at different concentrations(25-200 μmol/L)on oscillation frequency and amplitude were analyzed.Results MDMi cells with typical ramified morphology were successfully genera-ted,exhibiting high expression of microglia-specific markers IBA1 and TMEM119.These cells responded robustly to LPS stimulation,with approximately a fivefold increase in TNF-α secretion.Calcium imaging revealed active spontaneous cal-cium oscillations under resting conditions.Propofol modulated calcium activity in a dose-dependent manner:compared with the control group,50 μmol/L and 100 μmol/L propofol significantly increased both the frequency and amplitude of calcium oscillations,whereas 200 μmol/L propofol suppressed these parameters.Conclusion MDMi exhibit key pheno-typic and functional characteristics of human microglia.Propofol exerts bidirectional,concentration-dependent effects on calcium signaling in MDMi,enhancing oscillatory activity at clinically relevant concentrations while inhibiting it at higher concentrations.关键词
