摘要

Abstract

Agrilus zanthoxylum,a destructive wood-boring pest in the planting area of Zanthoxylum bungeanum in northern China.Due to its concealed harmful symptoms,physical control and single chemical application are far from effective,making it difficult to prevent and control.The use of host plant volatiles for the eco-friendly management of A.zanthoxylumi represents an innovative approach that can significantly diminish the environmental pollution associated with the widespread application of chemical insecticides.Chemosensory protein,as an important part of the olfactory system of in-sects,plays an important role in binding and transporting plant volatiles.Studying the binding charac-teristics of chemosensory proteins to host volatiles is an important step to develop new botanical at-tractant and repellents.In this study,fluorescent competitive binding experiments were conducted to analyze the binding characteristics between AzanCSP4(a chemosensory protein of Agrilus zanthoxy-lumii)and 25 host volatiles of Zanthoxylum bungeanum.The results showed that AzanCSP4 has a binding site number of 1.37 when interacting with the fluorescent probe N-phenyl-1-naphthylamine(1-NPN),and a Ki value of 6.02.AzanCSP4 was found to bind to six ligands,which included five al-kenes and one ester.It exhibited strong binding affinity with β-caryophyllene,β-pinene,terpinyl ace-tate and cubebene(0<Ki<10),while displaying weaker binding affinity for α-pinene and caryo-phyllene(10<Ki<20).These findings help our understanding of the molecular mechanisms by which AzanCSP4 recognizes the volatiles of Z.bungeanum,providing a theoretical foundation for the devel-opment of environmentally friendly control technologies for A.zanthoxylumi.
