α-葡萄糖苷酶抑制剂筛选方法的构建及应用
Establishment and application of a screening method for α-glucosidase inhibitors

Abstract
To rapidly discover lead compounds with α-glucosidase inhibitory activity from natural products for the treatment of type Ⅱ diabetes,this study established and validated an efficient screening platform based on ultraviolet spectrophotometry.Through systematic optimization,the optimal conditions were determined as follows:detection wavelength of 394 nm,200 μL of substrate(2 mg/mL),100 μL of enzyme(2 000 U/mL),100 μL of PBS buffer,a temperature of 37 ℃,and a reaction time of 300 s.Using this optimized system,the inhibitory activities of 206 aqueous extracts of traditional Chinese medicines(TCMs)were screened,with acarbose as the positive control.The results revealed that 28 TCM extracts exhibited inhibition rates exceeding 30%.Among them,the inhibition rates of Mulberry Bark(Morus alba L.),Silkworm Frass(Bombyx mori L.),and Honey-Processed Mulberry Bark all surpassed 60%,with IC50 values comparable to that of acarbose.This study demonstrates that the established system is straightforward,sensitive,and reliable,providing not only an experimental basis for the rapid screening of α-glucosidase inhibitors but also a feasible approach for discovering novel anti-diabetic active components from TCMs.关键词
