Abstract

An HPLC-RID method for determining the sucrose mass fraction in COVID-19 mRNA vaccines was established using an aminopropyl-bonded silica column(4.6 mm × 250 mm,5.0 μm)with an acetonitrile-water solution as the mobile phase.Optimization was performed on the mobile phase ratio,flow rate,and column temperature.The method demonstrated a good linear relationship within the sucrose mass concentration range of 5.05-30.30 mg/mL,along with satisfactory specificity,repeatability,accuracy,and durability.Both the limit of quantification(LOQ)and limit of detection(LOD)met the analytical requirements.This approach not only provides a reliable analytical means for quantifying sucrose in COVID-19 mRNA vaccines but also establishes a research foundation for sucrose determination in other mRNA-based vaccines.
