登革病毒2型感染对新型hTim4基因修饰小鼠脾的病理损伤
A novel hTim4 genetically modified mouse model reveals enhanced splenic pathogenesis upon dengue virus serotype 2 challenge

Abstract
Objective To investigate pathological changes in the spleens of human T cell immunoglobulin and mucin domain containing 4(hTim4)genetically modified mice(hTim4 mice)induced by serotype 2 dengue virus Tr.1751(DENV2).Methods The hTim4 mice were intracerebrally inoculated with DENV2(1× 103 PFU/mL).After inoculation,body mass was monitored,and deaths were documented.Viremia was recorded,along with viral load and pathological changes in the spleens of dying mice;vascular leakage was also determined.Results Mice inoculated with DENV2 exhibited continuous body mass reduction,all succumbing to infection.Significant vascular leakage in spleens,lungs,livers,and intestines was observed,along with severe spleen atrophy.Spleen white and red pulp boundaries became blurry,and cell counts decreased in both PALS and margin zones;apoptotic cells were scattered in white pulp,especially in the margins.Conclusions DENV2 infection caused splenic hemorrhage,cell reduction,and atrophy in hTim4 mice.关键词
