Interventional effect and preliminary mechanism exploration of Isaria felina on metabolic disorders in high-fat diet-induced obese mice

Abstract
Objective To investigate the interventional effect and underlying mechanism of Isaria felina(IF)on metabolic disorders in high-fat diet(HFD)-induced obese mice.Methods Thirty SPF grade male KM mice were stratified randomly by body mass into control,model,white kidney bean,50 mg/kg IF,and 100 mg/kg IF groups(n=6 mice per group).Control mice were fed a maintenance diet(10%fat energy),while mice in the other groups were fed a HFD(60%fat energy).Control and model mice received 0.9%sodium chloride solution daily by gavage,mice in the white kidney bean group received white kidney bean extract 50 mg/(kg·d)by gavage,and mice in 50 mg/kg and 100 mg/kg IF groups received 50 and 100 mg/(kg·d)IF by gavage,respectively.After continuous gavage for 6 weeks,an oral glucose tolerance test(OGTT)was performed and serum levels of total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),aspartate aminotransferase(AST),alanine aminotransferase(ALT),alkaline phosphatase(ALP),urea,and creatinine(CRE)were measured.Liver and adipose tissues were excised and weighed to calculate the liver index and fat index.Epididymal adipose tissue and liver tissue were stained with hematoxylin-eosin(HE)to observe histopathological changes and hepatic lipid accumulation was evaluated by Oil red O staining.Results Compared with the model group,body mass,fasting blood glucose,area under the OGTT curve,serum levels of TC,TG,LDL-C,abnormally elevated HDL-C,AST,ALT,ALP,Urea,and CRE,and liver and adipose tissue mass and indices were all significantly reduced in mice in the 50 mg/kg IF group and 100 mg/kg IF group(P<0.05).IF alleviated pathological adipocyte hypertrophy in epididymal adipose tissue,and mitigated hepatic steatosis and lipid accumulation.Conclusions IF can improve glucose intolerance,reduce serum lipids,inhibit body fat accumulation,alleviate hepatic steatosis,and mitigate hepatorenal dysfunction in HFD-induced obese mice.关键词
