Mapping critical epitope residues of antigenic site 4 on the AF72 vaccine strain of foot-and-mouth disease virus serotype A using porcine single B-cell neutralizing antibodies

Abstract
The antigenic structure of the AF72 vaccine strain of serotype A foot-and-mouth disease virus(FMDV)was characterized by neutralizing monoclonal antibodies(mAbs)derived from porcine single B cells.Twelve naturally paired immunoglobulin heavy-and light-chain sequences were identified and cloned into expression vectors,and recombinant antibodies were produced in HEK-293F cells.The reactivity and neutralizing activity of these mAbs were validated by indirect ELISA,indirect immunofluorescence assay,and virus neutralization tests.Furthermore,key amino acid residues of the antigenic epitopes were identified by screening escape mutants and confirmed using reverse genetics to rescue single-point mutant viruses.The results indicated that 11 of the 12 mAbs were specific to FMDV serotype A,among which six exhibited neutralizing activity against the AF72 strain.Specifically,residues VP3 A68(targeted by PAF26,PAF27,PAF31,and PAF32),VP3 G70(targeted by PAF42),and VP3 E139(targeted by PAF44)were identi-fied as critical recognition sites.These residues formed a spatially clustered region on the capsid surface,constituting the core epitope of antigenic site 4.This study refines the antigenic structural characteristics of the AF72 strain and supports the rational design of serotype A FMDV vaccines.关键词
