摘要

Abstract

This study aimed to investigate whether cell lines expressing foot-and-mouth disease virus(FMDV)neutralizing antibodies(NAbs)could inhibit viral infection and replication.First,plasmids expressing FMDV NAbs in the form of single-chain fragments variable(scFv)were constructed.To further enhance the secretion efficiency of NAbs into the extracellular space,four secretory signal peptides were added into each scFv.The expression vectors were transfected into BHK-21 cells via liposome-mediated transfect ion,fol lowed by puromycin selection to obtain the corresponding transgenic cell lines.Subsequently,FMDV replication in the transgenic cell lines was assessed using Western-blot and TCID50 assays.Results demonstrated that the mIg κ-signal peptide-tagged NAbs significantly inhibited FMDV replication in BHK-21 cells,and the degree of cytopathic effect induced by viral infection was markedly reduced.These findings indicate that scFv-formulated NAbs can potently suppress FMDV replication in vitro,offering novel insights for controlling livestock epidemics.
