Preparation and immunogenicity evaluation of mRNA vaccines targeting the canine parvovirus VP2 protein

Abstract
To design and prepare multiple mRNA vaccine candidates targeting the VP2 antigen of ca-nine parvovirus(CPV),and to evaluate their in vitro expression and immunogenicity in mice,the standard VP2 gene sequence was obtained from the NCBI database.Secretory and membrane-anchored VP2 constructs,designated as sVP2 and mVP2,were designed and inserted into an mRNA expression vector.Functional mRNA were synthesized through plasmid linearization,in vitro transcription,purification,and capping.Ex-pression of the target antigens was verified by Western-blot and indirect immunofluorescence assay.Lipid nanoparticle(LNP)-encapsulated mRNA vaccines(LNP-mRNA vaccines)were then prepared,and their immunogenicity was evaluated in mice following an established immunization protocol.The results demonstrated that two mRNA vaccine candidates were successfully constructed and efficiently expressed the VP2 antigen in cells.Notably,the sVP2 construct induced humoral immune responses and neutralizing antibody production in mice.This study provides experimental evidence and theoretical support for the development of mRNA vaccines against CPV.关键词
