摘要

Abstract

To explore the function of the ZmL-PO gene and to clarify the molecular regulatory mecha-nism of the increase in lysine(Lys)content in maize(Zea mays L.)kernels,using maize B73 as the material,the coding region of ZmL-PO was obtained through molecular cloning and its sequence was sequenced.Combined with bioinformatics analysis,the protein function of ZmL-PO was predicted,and the gene expression patterns in different tissues and at different stages of maize B73 were ana-lyzed.The results showed that the length of the CDS of the cloned ZmL-PO was 1 245 bp,and it was predicted to contain 415 amino acids with a molecular weight of approximately 43.94 kD and an isoe-lectric point of 5.79.Bioinformatics analysis indicated that ZmL-PO belonged to a hydrophilic protein located in the chloroplast,and its secondary structure was mainly α-helix.Systematic phylogenetic analysis showed that the evolutionary relationship between ZmL-POprotein and Sorghum bicolor was the closest.The qRT-PCR results showed that the expression level of ZmL-PO was the highest in the kernels 20 days after pollination.
