Establishment and optimization of a regeneration and rapid propagation system in Miscanthus sinensis

Abstract
Miscanthus sinensis is an ornamental energy grass possessing ecological restoration,soil conservation,and water retention capabilities.However,its production has traditionally relied on asexual reproduction,which is inefficient and seasonally constrained.Therefore,the establishment and optimization of a regeneration and rapid propagation system for M.sinensis using tissue culture technology offers considerable practical value.Using the seeds as extracts,we constructed an efficient and stable regeneration and rapid propagation system for M.sinensis.The results revealed that using manual debran or soaking in 50%sulfuric acid,followed by disinfection with HgCl2 for 15 min,significantly enhances seed germination rates and reduces contamination rates(P<0.05).During callus induction,the use of 8.0 mg·L-1 Dicamba resulted in an induction rate of 75.2%.Moreover,the addition of 1 000.0 mg·L-1 active carbon during subculture effectively mitigated callus browning.During callus differentiation,the use of 2.0 mg·L-1 zeatin significantly increased its differentiation rate(P<0.05),with a regeneration efficiency of 6.5.For the transplantation of M.sinensis seedlings,the base comprising peat soil,vermiculite,and perlite(1꞉1꞉1),yielded the highest survival rate(98.8%).Collectively,this system provides a technical foundation for the large-scale cultivation,germplasm conservation,and molecular breeding of M.sinensis.关键词
