紫色芹菜花青素合成酶AgANS基因克隆及不同光质条件的表达分析
Cloning of Anthocyanidin Synthase AgANS Gene and Its Expression Under Different Light Conditions in Purple Celery

Abstract
[Objective]The anthocyanidin synthase(ANS)gene of purple celery was investigated to provide a theoretical basis for studying the molecular mechanism of ANS gene in anthocyanidin synthesis of purple celery.[Method]The bioinformatics of AgANS gene obtained from the petioles of purple celery at 8-leaf stage by gene cloning was analyzed.The anthocyanidin content and expression level of AgANS gene were detected under different light quality treatments of red light,blue light and red-blue compound light with different proportion.[Result]The full length of cDNA sequence of AgANS gene was 1 278 bp with 425 amino acids and the acid amino acids accounted for 22.82%.The AgANS protein contained four conserved domains,and the secondary structure consisted of α-helices(35.76%),and random coils(43.76%).The evolution analysis showed that the AgANS protein from purple celery was closely related to that of Daucus carota(Apiaceae)and more distantly related to species from Vitaceae and Bombacaceae.The relative expression level of AgANS gene treated by blue light was 4.47 times that of CK.The red light,blue light and red-blue compound light all were beneficial to anthocyanin accumulation in petioles of purple celery and the anthocyanin content in petioles of purple celery was the highest under the blue light treatment.[Conclusion]The AgANS gene is successfully cloned from purple celery.The anthocyanin content in petioles of purple celery and expression level of AgANS gene are significantly influenced by light quality,and the blue light has the most significant effect on anthocyanin accumulation in petioles of purple celery.关键词
