Determination of Quercetin,Luteolin and Kaeampferol in Broccoli by N-doped Carbon Cloth Fiber-C18 Purification Combined with HPLC-MS/MS

Abstract
To improve the detection efficiency of quercetin,luteolin and kaempferol in broccoli by HPLC-MS/MS,self-prepared nitrogen-doped carbon cloth fiber(NACC)combined with C18 was adopted for adsorptive purification of matrix impurities.The type and dosage of adsorbents,purification temperature and solution pH were optimized,and the analytical performance of the method was evaluated.The results showed that the three target substances exhibited good linear relationships within the range of 50-5 000 μg·kg-1,with R2 ranging from 0.999 4 to 0.999 6.The limits of detection were 50,50 and 200 μg·kg-1,and the limits of quantification(LOQ)were 100,100 and 500 μg·kg-1,respectively.The average recoveries were 97.2％～99.2％,with relative standard deviation(RSDs)of 1.0％～1.3％.Single-factor experimental optimization confirmed the optimal purification conditions as follows:C18 dosage of 100 mg,NACC area of 2 cm2,purification temperature of 50 ℃,and solution pH of 6.Under these conditions,interfering components such as pigments could be effectively removed while ensuring high recoveries and satisfactory retention of target analytes.The proposed method presented favorable detection performance and recoveries in the analysis of practical broccoli samples.This study indicates that the established method features high purification efficiency,simple pretreatment and high detection sensitivity,which can provide a technical reference for the efficient determination of flavonoids in plant-based foods,and also expand the application potential of NACC materials in sample pre treatment.关键词
