Expression and clinical significance of serum latent transforming growth factor binding protein 2 and trefoil factor family peptide 3 in patients with pancreatic cancer

Abstract
Objective To analyze the expression and clinical significance of serum latent trans-forming growth factor-binding protein 2(LTBP2)and trefoil factor family peptide 3(TFF3)in pa-tients with pancreatic cancer.Methods A total of 146 patients with pancreatic cancer treated at Handan Central Hospital from February 2023 to October 2024 were selected as the pancreatic cancer group,and 70 patients with benign pancreatic tumors in the same period were selected as control group.The serum levels of LTBP2 and TFF3 were detected by the enzyme-linked immunosorbent assay(ELISA)method,and the relationships of serum LTBP2 and TFF3 levels with clinicopathological parameters were analyzed.Pearson method was used to analyze the correlation between serum LTBP2 and TFF3 expression levels in patients with pancreatic cancer.Univariate and multivariate Logistic regression analyses were employed to identify factors influencing the occurrence of pancreatic cancer.The diag-nostic efficacy of serum LTBP2 and TFF3(both individually and their combination)for pancreatic cancer was evaluated by the receiver operating characteristic(ROC)curve.Results The serum expression levels of LTBP2 and TFF3 in the pancreatic cancer group were significantly higher than those in the control group(P＜0.001).A positive correlation was observed between serum LTBP2 and TFF3 expression(r=0.505,P＜0.05).LTBP2 and TFF3 were significant influencing factors for the occurrence of pancreatic cancer(P＜0.05).For every 1 μg/mL increase in LTBP2 expres-sion level,the risk of pancreatic cancer increased by 20.50％;for every 1 ng/mL increase in TFF3 expression level,the risk of pancreatic cancer increased by 33.80％.The areas under the curve(AUCs)for the individual diagnosis of pancreatic cancer by serum LTBP2 and TFF3 were 0.831(95％CI,0.774 to 0.878)and 0.953(95％CI,0.916 to 0.977),respectively.The AUC for combined diagnosis was 0.971(95％CI,0.939 to 0.989),with a sensitivity of 91.78％and a specificity of 98.57％.Combined diagnosis was superior to individual diagnosis(Zcombined-LTBP2=5.298,Zcombined-TFF3=2.113,P＜0.05).LTBP2 and TFF3 expression were correlated with nerve invasion,TNM stage,and lymph node metastasis(P＜0.05).Conclusion Serum LTBP2 and TFF3 levels are significantly upregulated in patients with pancreatic cancer and are closely associated with nerve invasion,TNM stage,and lymph node metastasis.Combined detection exhibits high di-agnostic accuracy and holds promise as important biomarkers for the clinical management of pancre-atic cancer.关键词
