理气活血滴丸对低血流剪切力诱导的人脐静脉内皮细胞凋亡及炎性反应的影响
Effect of Liqi Huoxue Dropping Pills(理气活血滴丸)on Apoptosis and Inflammatory Response of Endothelial Cells Induced by Low Blood Flow Shear

Abstract
Objective To investigate the effects of Liqi Huoxue Dropping Pills(理气活血滴丸)on the apoptosis of human um-bilical vein endothelial cells and the expressions of inflammation-related factors induced by low blood flow shear force.Methods A total of 60 healthy male SD rats were randomly divided into model group and low,medium and high dose groups of Liqi Huoxue Dropping Pills,with 15 rats in each group.The low,medium and high dose groups of Liqi Huoxue Dropping Pills were giventhe drug with the corresponding doses[43.7,87.5,175 mg/(kg·d)]by gavage once every 12 hours.The model group was given 0.9％sodium chloride solution[10 mL/(kg·d)]by gavage every 12 h.All rats were given the drug continuously for 96 h.The blood was collected from the abdominal aorta 1 h after the last dose,and the drug serum was routinely prepared.Human ompha-lovenous endothelial cells(HUVECs)cultured in vitro were divided into blank group,model group and Liqi Huoxue Dropping Pills low,medium and high dose groups.Except the blank group,the other groups were cultured in the medium containing the corre-sponding drug serum for 48 h,and then loaded with low fluid shear force(4 dynes/cm2)for 24 h.The apoptosis of HUVECs in dif-ferent groups was detected by TUNEL method.The endothelial cell adhesion factor 1(VCAM-1),E-selectin,interleukin-1 β(IL-1 β)and the expressions of apoptosis-related factors cysteinyl aspartate-specific proteinase 3(Caspase-3)and B cell lymphoma-2 gene(bcl-2)in different HUVECs were detected by Western blotting.Results Compared with those in the blank group,the apoptosis rate and protein expressions of VCAM-1,E-selectin,IL-1 β and Caspase-3 in the model group were sig-nificantly increased,while bcl-2 protein expression was decreased,with statistical significance(P＜0.05).Compared with those in the model group,the apoptosis rate and protein expressions of VCAM-1,E-selectin,IL-1 β and Caspase-3 were lower in the Liqi Huoxue dropping Pills groups,while bcl-2 protein expression was increased,and the differences were statistically signifi-cant(P＜0.05).Conclusion Liqi Huoxue Dropping Pills can inhibit the apoptosis of HUVECs under low fluid shear load,which may be related to down-regulating inflammatory factors and inhibiting inflammatory damage.关键词
