固本止咳平喘颗粒对咳嗽变异性哮喘大鼠NGF/TrkA信号通路的影响
Effect of Guben Zhike Pingchuan Granule(固本止咳平喘颗粒)on NGF/TrkA Signaling Pathway in Cough Variant Asthma Rats

Abstract
Objective This study was to investigate the effects of Guben Zhike Pingchuan Granule(固本止咳平喘颗粒)on the nerve growth factor(NGF)/tyrosine kinase A(TrkA)signaling pathway in the rats with cough variant asthma(CVA)and to explore the mechanism by which Guben Zhike Pingchuan Granule improves CVA neuroinflammation.Methods SD rats were ran-domly divided into seven groups,including normal group,model group,montelukast sodium group,Suhuang Zhike Capsule(苏黄止咳胶囊)group and Guben Zhike Pingchuan Granule low,medium and high dose groups.The CVA model was established by smoke exposure combined with ovalbumin(OVA)sensitization.The drug-treated groups were given oral administration of the drugs for 15 days after the model was established.After drug administration,the left lung was perfused with bronchoalveolar lavage fluid(BALF),and the contents of NGF,interleukin-13(IL-13)and leukotriene E4(LTE-4)in the BALF were detected by ELISA.The pathological changes in the right upper lobe of the lung were observed by HE staining.The protein expressions of NGF and TrkA in the lower lobe of the right lung were detected by Western blot.Results The ELISA results showed that compared with those in the model group,the NGF levels in BALF of the rats in the low,medium and high dose Guben Zhike Pingchuan Granule groups and the Suhuang Zhike Capsule group were significantly decreased(P＜0.01).The IL-13 levels of the medium and high dose Guben Zhike Pingchuan Granule groups and the montelukast sodium group were significantly decreased(P＜0.05 or P＜0.01)and the LTE-4 levels of the montelukast sodium group,Suhuang Zhike Capsule group and low,medium and high dose Guben Zhike Pingchuan Granule groups were significantly decreased(P＜0.01).The HE staining results suggested that the Guben Zhike Pingchuan Granule low,medium and high dose groups can effectively reduce the thickening of alveolar septa and al-leviate the infiltration of inflammatory cells.The Western Blot detection results suggested that compared with those in the model group,the expressions of NGF and TrkA in the Guben Zhike Pingchuan Granule high dose group was significantly down-regula-ted(P＜0.01),the expression of NGF in the Guben Zhike Pingchuan Granule middle dose group was significantly down-regula-ted(P＜0.01)and the expression of TrkA in the montelukast sodium group was down-regulated(P＜0.05).Conclusion Guben Zhike Pingchuan Granule can effectively alleviate the excessive infiltration of lung inflammatory cells in CVA rats,and this effect may be related to the regulation of the NGF/TrkA signaling pathway.关键词
