Recent advances in phosphocreatine metabolism and its role in cancer research

Abstract
Phosphocreatine(PCr),traditionally recognized as an intracellular high-energy phosphate compound,mainly functions as a rapid energy buffer through the creatine kinase system to meet the high-energy demands of cells.However,recent studies have revealed that the role of PCr extends beyond this conventional view;it plays multiple roles in tumor metabolism,especially in maintaining tumor stem cell stemness,regulating epigenetics,remodeling metabolism,and contributing to drug resistance.PCr not only supports the energy supply required for the rapid proliferation of tumor cells but also affects processes such as chromosome segregation and cell cycle regulation by modulating the stability of key proteins.Moreover,PCr metabolism can serve as a therapeutic target,where inhibiting its synthesis pathway significantly suppresses tumor growth and exhibits synergistic effects with existing epigenetic inhibitors,providing new strategies for cancer therapy.This review focuses on the latest advancements including the regulation of tumor cell proliferation,migration,and invasion by PCr,its core role in maintaining tumor stemness and epigenetic remodeling,its involvement in the molecular pathways of tumor drug resistance,and new strategies for anti-tumor therapy targeting PCr metabolism.This review systematically summarizes the recent progress in PCr metabolism in tumorigenesis,tumor progression,and treatment,analyzes its multifaceted functions in tumor cell biology,and explores its potential application value in targeted therapy.关键词
