二甲双胍治疗神经病理性疼痛作用机制研究进展
Research progress in mechanisms of metformin against neuropathic pain
摘要
Abstract
Neuropathic pain(NP),also known as neurogenic pain,is a form of chronic pain caused by damage to or disease of the somatosensory nervous system.Research suggests that metformin,a classic antidiabetic drug,holds therapeutic potential in the field of pain management.However,its specific targets and the interactive network between these multiple targets remain unclear.This paper reviews the latest research findings about how this drug alleviates NP through adenosine monophosphate-acti-vated protein kinase(AMPK),downstream AMPK signalling molecules and their interaction networks,and the mammalian target of rapamycin complex 1(mTORC1).Based on analysis of the mechanisms and variations in therapeutic efficacy,the advantages of and challenges to metformin in the treatment of NP are summarized while a biomarker-based,stratified precision analgesic strategy is recommended,which is expected to provide a framework for optimizing dosing regimens,developing targeted interven-tions for NP,and implementing personalized pain management.关键词
