Abstract

The NRT2 family of high-affinity nitrate transporters plays a crucial role in nitrogen uptake and redistribution under nitrogen-deficient conditions.In this study,30 different potato varieties were analyzed to investigate the expression patterns of StNRT2.5 using quantitative real-time PCR(qRT-PCR),and bioinformatics analysis of its conserved domains,promoter elements,and phylogenetic relationships.To further verify the function of StNRT2.5,protein-protein interaction prediction and the expression analysis of interacting proteins were conducted using the STRING database.The results showed significant variation in StNRT2.5 expression among different potato varieties and tissues.StNRT2.5 contains conserved domains characteristic of nitrate transporter family proteins.Promoter analysis revealed the presence of multiple cis-acting elements,including light-responsive elements,hormone-responsive elements,as well as promoter and enhancer elements.Phylogenetic analysis indicated high homology between StNRT2.5 and primitive cultivated diploid potato and Solanum verrucosum.Expression analysis of interacting proteins showed that StNR was upregulated in leaves,while StNRT3.1 was predominantly expressed in roots.
