回肠黏膜非编码RNA与猪剩余采食量社会遗传效应的关联研究
Association of ileal mucosal non-coding RNA with social genetic effects of residual feed intake in pigs

Abstract
This study aimed to investigate the differential expression characteristics of non-coding RNA(ncRNA)in the ileal mucosa under different residual feed intake-social genetic effects(RFI-SGE).The study involved 204 healthy Duroc sows,with feeding behavior measured during the 30-115 kg finishing period using an electronic feeding record system.Based on RFI-SGE values,high(HRS)and low(LRS)RFI-SGE groups(10 sows each)were constructed,and four sows with extreme SGE phenotypic values from each group were chosen to collect ileal mucosal tissue for transcriptomic sequencing.Biological information analysis was used to identify differentially expressed ncRNAs and construct a competitive endogenous RNA(ceRNA)regulatory network.A total of 11 687 long non-coding RNAs(lncRNAs)and 577 microRNAs(miRNAs)were identified in the ileal mucosa,among which 1 117 ncRNAs showed significant differential expression(956 up-regulated lncRNAs and 98 down-regulated lncRNAs;35 up-regulated miRNAs and 28 down-regulated miRNAs).The target genes of differentially expressed miRNAs were significantly enriched in cell mitosis,modification-dependent protein binding,and sulfur metabolism pathways;the target genes of differentially expressed lncRNAs were primarily involved in fat digestion and absorption,amino acid digestion,and cholesterol metabolism.Through ceRNA network analysis,it was found that 15 up-regulated messenger RNAs(mRNAs)interacted with seven down-regulated lncRNAs,and seven up-regulated mRNAs,six up-regulated lncRNAs,and four down-regulated miRNAs formed a triple regulatory network.Among them,key lncRNAs such as LOC110261770,LOC102160805,and BGIG9823_46847 were significantly associated with genes related to amino acid metabolism.This study revealed the expression characteristics and functional networks of ileal mucosal ncRNAs in RFI-SGE,suggesting that ncRNAs may influence RFI-SGE by regulating cellular processes such as cell growth,amino acid metabolism,and lipid metabolism.The findings provide new candidate targets and a theoretical basis for molecular breeding in pig RFI-SGE,meanwhile opening up a new research perspective for improving feed efficiency in pigs.关键词
