Study on the in vitro Antibacterial Activity of Galla Chinensis,Catechu,and Coptis chinensis against Stenotrophomonas maltophilia

Abstract
Objective To systematically evaluate and compare the in vitro antibacterial activity of three traditional Chinese medicines(TCMs),Galla Chinensis,Catechu,and Coptis chinensis,against clinical isolates of Stenotrophomonas maltophilia.Methods The agar dilution method was employed to determine the minimum inhibitory concentration(MIC),MIC required to inhibit 50%(MIC50)and 90%(MIC90)of Galla Chinensis,Catechu,and Coptis chinensis against 150 clinical isolates;the minimum bactericidal concentration(MBC)was determined using the tube dilution method;time-kill curves were constructed using the plate count method;the Mann-Whitney U test was employed to compare MIC values across groups based on gender,age,specimen type,and drug susceptibility phenotype.Results All three TCMs exhibited significant in vitro antibacterial activity.Galla Chinensis demonstrated an MIC50 of 1.022 mg/mL and an MIC90 of 2.044 mg/mL,Coptis chinensis exhibited an MIC50 0.664 mg/mL and an MIC90 of 1.062 mg/mL,while Catechu showed an MIC50 8.278 mg/mL and an MIC90 13.249 mg/mL.The MBC ranges for Galla Chinensis,Catechu and Coptis chinensis were 2.555-3.861 mg/mL,1.660-2.725 mg/mL,and 20.695-28.278 mg/mL,respectively.Time-kill curves revealed distinct patterns:Galla Chinensis showed potent,delayed bactericidal activity,Coptis chinensis exhibited a sustained bacteriostatic effect,while Catechu demonstrated rapid bactericidal activity.No significant differences in MIC values were observed among groups stratified by gender,age,or specimen type,except for the differences in MIC values of Catechu among the antimicrobial susceptibility phenotype groups(P<0.05).Conclusion Galla Chinensis,Catechu and Coptis chinensis all possess effective in vitro inhibitory activity against Stenotrophomonas maltophilia.Among these,Galla Chinensis demonstrated the strongest efficacy and a distinct bactericidal kinetic profile,positioning it as the most promising candidate for further development.关键词
