酸枣仁知母配方颗粒黄酮类成分的HPLC含量测定及比较
Determination and Comparison of Flavonoids in Suanzaoren Formula Granules and Zhimu Formula Granules by High Performance Liquid Chromatography

Abstract
Objective To determine and compare the contents of flavonoid components including spinosin and mangiferin in Suanzaoren formula granules and Zhimu formula granules produced by manufacturers in Guangdong and Shenzhen before the implementation of national standards.Methods The content of spinosin in Suanzaoren formula granules and mangiferin in Zhimu formula granules was determined respectively by high performance liquid chromatography(HPLC).The HPLC method was performed on a Kromasil C18 column(4.6 mm×250 mm,5 µm)using gradient elution of acetonitrile-1%acetic acid solution as the mobile phase.The flow rate was 1.0 mL/min,the column temperature was maintained at 30℃,and the detection wavelength was set at 335 nm.Results The linear ranges of spinosin,mangiferin were 0.085 6-0.513 7 µg(r=0.999 999 9)and 0.078 56-0.392 8 µg(r=0.999 93),respectively.The highest content of spinosin was 1.55 times that of the lowest content in Suanzaoren formula granules produced by the two plants,but the highest content of mangiferin in Zhimu formula granules was 10.08 times that of the lowest content.Converting to the equivalent of raw herbs,the content of spinosin was all under the lower limit of national standards,while the content of mangiferin was under the lower limit of national standards in in two batches.Conclusion The quality of Suanzaoren formula granules had no obvious difference in Guangdong and Shenzhen manufacturers before the implementation of national standards,but there was significant distinction of Zhimu formula granules.The enforcement of national standards would propel quality enhancement in Chinese herbal formula granules.The dosage of national standard formula granules should be rationally administered in clinical practice.关键词
