溪黄草醇提物抑制血管生成的实验研究
Experimental study of the ethanol extract of Rabdosia serra for inhibiting angiogenesis

Abstract
Objective:To investigate the inhibitory effect of the ethanol extract of Rabdosia serra on angiogenesis.Methods:The ethanol extract of Rabdosia serra was prepared by reflux extraction with 80%ethanol.The 3-(4,5-di-methylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay was used to evaluate its inhibitory effect on hu-man umbilical vein endothelial cells(HUVECs).A cell scratch assay was performed to assess the extract's inhibition of HUVECs migration.Sorafenib served as the positive control.Western blot(WB)analysis was conducted to determine the effects of different concentrations of the extract on the expression of vascular endothelial growth factor(VEGF)and matrix metalloproteinase-9(MMP-9)in HUVECs.Enzyme-linked immunosorbent assay(ELISA)was employed to measure the extract's effect on the secretion of MMP-9 and VEGF.Reverse transcription-polymerase chain reaction(RT-PCR)was carried out to detect the effects of the extract on the mRNA expression of vascular endothelial growth factor A(VEGFA)and MMP-9 in HUVECs.A chick chorioallantoic membrane(CAM)model was established,with sorafenib as the positive control,to observe the anti-angiogenic effects of different concentrations of the ethanol extract.Results:The ethanol extract of Rabdosia serra inhibited HUVECs proliferation in vitro,with a half-maximal inhibitory concentration(IC₅₀)of 0.693 mg/ml at 24 h and 0.532 mg/ml at 48 h.Compared with the blank control group,the ex-tract significantly suppressed HUVECs migration(P<0.05),reduced the expression of VEGF and MMP-9,inhibited the secretion of VEGF and MMP-9,and downregulated VEGFA mRNA expression.Relative to the blank control group,sorafenib and the medium-and high-concentration groups of the ethanol extract significantly inhibited second-and third-order blood vessels in the CAM model(P<0.05).Conclusion:The ethanol extract of Rabdosia serra can inhibit microvascular angiogenesis.Its mechanism of action may involve the suppression of vascular endothelial cell migration and the downregulation of VEGF and MMP-9 expression.关键词
