Study on the Verification of the"Meridian Transplantation"Hypothesis and Acupuncture Effect Based on a Rabbit Autologous Renal Transplantation Model
摘要
Abstract
Objective:To provide experimental evidence for the"meridian transplantation"hypothesis by comparing the therapeutic effects of acupuncture at acupoints of different meridians on acute kidney injury(AKI)after autologous renal transplantation in rabbits. Methods:Adult female rabbits were randomly divided into a control group,a left spleen meridian group,a left kidney meridian group, and a right spleen meridian group, with 10 rabbits in each group.A rabbit model of ischemia-reperfusion injury after autologous ectopic renal transplantation was established as follows:bilateral kidneys and renal vessels were dissected sequentially;the renal artery was clamped for 50 minutes followed by reperfusion for 1 hour;the right kidney was resected for biopsy. The dissected left kidney with vascular pedicle was fixed to the inner abdominal wall along the nipple line(the course region of the Spleen Meridian of Foot-Taiyin).Renal injury was evaluated using the Paller tubular injury score.Modeling was considered successful if the Paller score was≥300 and serum creatinine(Scr)was<300 μmol/L.After modeling,no special treatment was given to the control group, while acupuncture was performed immediately after surgery in the other groups. The left spleen meridian group and right spleen meridian group received acupuncture at ipsilateral Spleen Meridian of Foot-Taiyin acupoints and"Zusanli"(ST 36)respectively;the left kidney meridian group received acupuncture at ipsilateral Kidney Meridian of Foot-Shaoyin acupoints and"Shenshu"(BL 23).All groups received electroacupuncture with dense-disperse waves,15 minutes per session, twice daily.Scr levels were measured before modeling, immediately after modeling, and on day 5 of treatment. On day 5 of treatment, animals were sacrificed and left renal biopsies were obtained for Paller scoring and intergroup comparison. Results:All model preparation met the evaluation criteria.Dynamic monitoring of Scr showed that Scr levels in the left spleen meridian group and left kidney meridian group on day 5 of treatment were significantly lower than those immediately after modeling(P<0.05),whereas no significant difference was found in the control group and right spleen meridian group(P>0.05).Comparison of Paller scores between the right kidney(non-ectopic after ischemia-reperfusion)and left kidney(ectopically transplanted to the spleen meridian region)revealed significantly lower scores in the left kidney(P<0.01).Immediately after modeling, there were no significant differences in Scr and right kidney Paller scores among groups(P>0.05).On day 5 of treatment, the overall difference in Scr among groups was statistically significant(P<0.05).Pairwise comparisons showed no significant difference between the left spleen meridian group and left kidney meridian group(P>0.05),but both had significantly lower Scr levels than the control group and right spleen meridian group(P<0.05).There was no significant difference between the control group and right spleen meridian group(P>0.05).The changing trend of Paller scores in the left kidney was consistent with Scr results(P<0.05).Conclusion:After autologous kidney transplantation to the course region of the spleen meridian, acupuncture at acupoints in this region(spleen meridian)yields renoprotective effects comparable to those of acupuncture at the original meridian(kidney meridian)acupoints. This suggests that after ectopic transplantation of the kidney, its functional regulation may shift to the new meridian along with its anatomical location, providing experimental evidence for the existence of the"meridian transplantation"phenomenon.关键词
