生肌玉红膏促进家兔全层皮肤缺损愈合的实验
Experimental Study on the Effect of Shengji Yuhong Gao(生肌玉红膏)on the Healing of Full-Thickness Skin Defects in Rabbits

Abstract
Objective:To investigate the effect and mechanism of Shengji Yuhong Gao in promoting wound healing in rabbit model of mechanical injury caused by full-thickness excision of dorsal skin.Methods:A total of 24 ordinary experimental rabbits were selected and randomly divided into four groups,with six rabbits in each group.All rabbits were used to establish models of mechanical injury caused by full-thickness excision of dorsal skin.The four groups were the blank control group(wound irrigation with normal saline and bandaging),the Kangfuxin liquid group(1.0 mL/kg),the low-dose Shengji Yuhong Gao group(0.3 g/kg),and the high-dose Shengji Yuhong Gao group(1.5 g/kg).Wound debridement and dressing change were performed once daily until wound healing.The wound healing time and wound healing rates on day 3,day 7,and day 14 were compared among the groups.Immunohistochemical CD34 staining was used to observe microvessel density in wound tissues,and HE staining was used to observe the number of fibroblasts in wound tissues.RT-qPCR was used to detect transforming growth factor-β(TGF-β1)mRNA expression in wound tissues,Western blotting was used to detect TGF-β1 protein expression in wound tissues,and the acid hydrolysis method was used to measure hydroxyproline(HYP)level in wound tissues.Results:Wound healing time in the treatment groups(Kangfuxin liquid group,low-dose Shengji Yuhong Gao group,and high-dose Shengji Yuhong Gao group)was shorter than that in the blank control group(P<0.05),and the high-dose Shengji Yuhong Gao group showed shorter healing time than low-dose Shengji Yuhong Gao group(P<0.05).At the same time points(day 3,day 7,and day 14),wound healing rates in the treatment groups were higher than those in the blank control group(P<0.05),and the high-dose Shengji Yuhong Gao group showed higher wound healing rate than low-dose Shengji Yuhong Gao group(P<0.05).Compared with day 3,there were increases in microvessel density,number of fibroblasts,TGF-β1 mRNA expression,TGF-β1 protein expression,and HYP level in wound tissues of each group on day 7(P<0.05).At the same time points(day 3 and day 7),all indicators in the treatment groups were higher than those in the blank control group(P<0.05),and the high-dose Shengji Yuhong Gao group showed higher values than low-dose Shengji Yuhong Gao group(P<0.05).Compared with day 7,there were increases in all indicators in the treatment groups on day 14(P<0.05),and the treatment groups showed lower values than blank control group(P<0.05).The high-dose Shengji Yuhong Gao group showed lower values than low-dose Shengji Yuhong Gao group(P<0.05).Conclusion:Shengji Yuhong Gao can promote wound healing in rabbit model of mechanical injury caused by full-thickness excision of dorsal skin.Its mechanism may be asso-ciated with increasing microvessel density and the number of fibroblasts in wound tissues and promoting granu-lation tissue growth via up-regulating TGF-β1 and HYP expression.关键词
