Effects of α-Linolenic Acid on the Transcriptome of Porcine Cumulus-Oocyte Complexes

Abstract
To investigate the mechanisms by which alpha-linolenic acid(ALA)promotes meiotic matu-ration and cumulus-oocyte interactions in porcine oocytes,this study employed an experimental design in which ALA was added to the culture medium of cumulus-oocyte complexes(COCs),with a non-supple-mented group serving as the control.Cumulus cells and oocytes were collected separately for transcriptome sequencing analysis,with three biological replicates per group.Differentially expressed genes(DEGs)were identified using DESeq2,followed by GO and KEGG enrichment analyses,clustering,and protein-protein interaction network analysis.The results revealed that ALA treatment led to the significant upregulation of 42 genes and downregulation of 38 genes in oocytes(P≤0.05).In granulosa cells,37 genes were signifi-cantly upregulated and 100 downregulated following ALA treatment(P≤0.05).In granulosa cells,DEGs were primarily enriched in pathways related to glycolysis/glucose transport and lipid metabolism,whereas in oocytes,DEGs were associated with actin filament organization and the extracellular space.Interaction analysis of key regulatory genes involved in pathways commonly enriched in both cumulus cells and oocytes highlighted a high frequency of interactions among genes in the cAMP signaling pathway.These findings provide essential foundational data for further exploration of the mechanisms by which ALA facilitates in vitro oocyte maturation and modulates cumulus-oocyte communication.关键词
