摘要

Abstract

To observe how different exercise modalities affect autophagic activity and inflammatory status in visceral adipose tissue(VAT)of obese mice,and to clarify the autophagy-inflammation interplay after exercise so as to uncover potential molecular mechanisms of exercise-induced fat loss,170 4-week-old male C57BL/6 mice were fed a high-fat diet(HFD)to establish obesity.Forty-two mice that reached the obesity criterion were randomly divided into a high-fat diet sedentary control(HHC group,n=9),a normal-diet sedentary(HNC group,n=9),a normal-diet plus endurance exer-cise(HNE group,n=12),and a normal-diet plus resistance exercise(HNR group,n=12).HNE and HNR mice com-pleted 8 week of treadmill running or ladder climbing,respectively.VAT was then harvested.Autophagy-and inflamma-tion-related mRNAs and proteins were quantified by RT-PCR and Western blotting;autophagosomes and protein localiza-tion were visualized by transmission electron microscopy and immunohistochemistry.The results showed that:① Com-pared with the HHC group,the expression of Atg5 mRNA and the LC3 Ⅱ/LC3 Ⅰ ratio in the HNC group were signifi-cantly decreased,the expression of ULK1,Becn1,Atg5 mRNA in the HNE group was significantly decreased,the ex-pression of Atg7,p62 mRNA was significantly increased,the expression of Becn1 protein and the LC3 Ⅱ/LC3 Ⅰ ratio was significantly decreased,and the expression of p62 protein was significantly increased.In the HNR group,the expression of Atg5 mRNA was significantly decreased,the expression of Atg7,p62 mRNA was significantly increased,the expres-sion of Becn1 protein and the LC3 Ⅱ/LC3 Ⅰ ratio was significantly decreased;compared with the HNC group,the ex-pression of Atg7,p62 mRNA in the HNE group was significantly increased,the expression of p62 protein was signifi-cantly increased,and in the HNR group,the expression of Atg7 mRNA was significantly increased;compared with the HNE group,the expression of Becn1 mRNA in the HNR group was significantly increased,and the expression of p62 protein was significantly decreased.② Compared with the HHC group,the expression of IL-1β protein in the HNC group was significantly decreased,the expression of IL-10 mRNA,IL-6,IL-10 protein in the HNE group was significantly in-creased,the expression of IL-1β protein was significantly decreased,and the expression of IL-6,IL-10 mRNA and pro-tein in the HNR group was significantly increased,the expression of IL-1β protein was significantly decreased;compared with the HNC group,the expression of IL-10 mRNA in the HNE group was significantly increased,the expression of IL-6,IL-10 protein was significantly increased,the expression of IL-1β protein was significantly decreased,and the expression of IL-6,IL-10 mRNA and protein in the HNR group was significantly increased,the expression of IL-1β protein was sig-nificantly decreased;compared with the HNE group,the expression of IL-6 mRNA in the HNR group was significantly increased,and the expression of IL-10 mRNA was significantly decreased.③ p62 showed a moderate correlation with IL-10 and IL-1β at the gene level(r=0.57*and-0.69*),and the expression of p62 protein also showed a moderate correlation with IL-6 and IL-10,with correlation coefficients reaching 0.51*and 0.64*,respectively.The expression of LC3 protein showed a good correlation with IL-10 protein expression(r=0.55*,P<0.05),and ULK1,Atg5,Atg7 genes all showed a good correlation with IL-10(-0.43,-0.67*,-0.62*).
