Countercurrent extraction process optimization and kinetic study of 10-DABⅢ and Taxol from Taxus yunnanensis

Abstract
To optimize the mechanical agitation-assisted countercurrent extraction process for 10-deacetylbaccatin Ⅲ(10-DABⅢ)and Taxol from Taxus yunnanensis branches and leaves,this study investigated the impact of solvent ratio on mass transfer in countercurrent extraction based on non-dissolution release kinetics theory.Using the extraction rate of 10-DABⅢ and Taxol as the evaluation index,single-factor experiments were conducted with three factors:extraction time,temperature and feed liquid ratio.The L9(34)orthogonal experiment was employed to comprehensively evaluate and optimize the four-stage mechanical agitation-assisted countercurrent extraction process parameters.A kinetic model was established using Fick's first law for solvent ratio[ln(M-H)]to describe the concentration change pattern[ln(C1-C0)]before and after single-stage extraction during countercurrent extraction.The model feasibility was validated through three extraction methods:mechanical agitation-assisted countercurrent extraction,oscillation-assisted countercurrent ex-traction,and percolation countercurrent extraction.The results showed that:① Extraction feed liquid ratio and tempera-ture were the primary factors affecting the extraction rate of 10-DABⅢ from T.yunnanensi using four-stage mechanical agitation-assisted countercurrent extraction.In all three extraction methods,mechanical agitation,oscillation and percola-tion,the ln(M-H)showed a good linear relationship with the ln(C1-C0)before and after single-stage extraction.② The optimized four-stage mechanical stirring-assisted countercurrent extraction process achieved an extraction temperature of 50℃,a solid-to-liquid ratio of 1∶5,and a processing time of 6 minutes,yielding 95.47%and 98.54%extraction rates for 10-DABⅢ and Taxol respectively.③ The ln(C1-C0)and ln(M-H)values in the three countercurrent extraction methods exhibited linear correlations,with the model showing excellent agreement with experimental results.In conclu-sion,the established four-stage mechanical stirring-assisted countercurrent extraction process provides valuable reference for both experimental design and industrial-scale production planning of plant component extraction.关键词
