黄瓜CsHCT基因家族全基因组鉴定及低温胁迫表达分析
Genome-wide identification and low temperature stress response expression analysis of the cucumber CsHCT gene family

Abstract
Hydroxycinnamoyl transferase(HCT),a member of the BAHD acyltransferase family,plays a key role in regulating the synthesis of lignin and chlorogenic acid.It is widely involved in plant growth and development,and respon-ses to abiotic stresses.In this study,based on the whole genome information of cucumber,bioinformatics methods was used to identify and analyze the CsHCT family members,and investigate the physical and chemical properties,chromo-some distribution,gene structure,promoter cis-acting elements and expression character-istics under cold stress,tissue expression profile and abscisic acid(ABA)treatment response of CsHCT42 by quantitative real-time PCR(qRT-PCR).The results indicated that a total of 59 members of the CsHCT family were identified,encoding 224-498 amino acids and evenly distributed across seven chromosomes.Theoretical isoelectric points ranging from 4.48 to 9.30,most CsHCT gene family members were unstable and hydrophobic.Phylogenetic tree analysis divided CsHCT members into 5 subcla-des,with genes in the same subclade sharing similar gene structures and conserved domains;Collinearity analysis indica-ted that some genes in cucumber,Arabidopsis and rice may have similar functions.Subcellular localization predictions in-dicated that CsHCT proteins were mainly located in the cytoplasm.The analysis of the promoter cis-acting elements showed that the promoter regions of CsHCT contained many cis-acting elements related to plant growth and development,abiotic stress responses,and hormone responses.Additionally,transcriptomic expression analysis revealed that CsHCT genes varied in response to low-temperature stresses,with significant expression changes over time.qRT-PCR results showed that the gene expression,such as CsHCT42 and CsHCT44 under low-temperature stress were consistent with the transcriptome data.The expression of CsHCT42 was tissue-specific and induced expression of CsHCT42 in ABA treat-ment.The findings of this study lay a foundation for elucidating the cold tolerance mechanism of cucumber and breeding new cucumber varieties with low-temperature tolerance.关键词
