Establishment of A Microspore Culture System and Development of Clubroot-Resistant DH Germplasm in Brassica juncea var.napiformis

Abstract
[Objective]This study aimed to establish an efficient microspore culture system for tuberous mustard(Brassica juncea var.napiformis)and to create breeding materials resistant to clubroot disease.[Method]Key steps including microspore developmental stage,embryoid induction,plant regenera-tion,and ploidy identification were optimized,and the regenerated DH(doubled haploid)populations were evaluated for disease resistance.[Result]The optimal microspore developmental stage for culture corresponded to bud lengths of 2.0-4.0 mm,at which the microspores were at the late uninucleate stage.Following induction with heat shock at 33 ℃ for 2 days,plant regeneration was achieved on the opti-mized medium.Flow cytometry analysis revealed a spontaneous diploidization rate of 9.57%,and three resistant lines were screened from 26 DH lines.[Conclusion]A complete technical procedure from mi-crospore culture to disease resistance evaluation was established for tuberous mustard,confirming its practical value in the rapid development of homozygous breeding materials.关键词
