黄金芽茶树光敏色素家族基因鉴定与表达分析
Identification and Expression Analysis of Phytochrome Family Genes in Camellia sinensis cv.Huangjinya

Abstract
Camellia sinensis cv.Huangjinya has attracted significant attention for its high ornamental val-ue due to the characteristic yellowing of its new shoots during three seasons.Red light significantly affects the leaf color transition from yellow to green in Huangjinya,and phytochromes(PHY)are the receptors that per-ceive and transmit red light signals.In this study,five CsPHY genes of Huangjinya were screened from tea tree genome database and successfully cloned,which were named CsPHYA1,CsPHYA2,CsPHYB1,CsPHYB2 and CsPHYE.Bioinformatics analysis,phylogenetic tree analysis,and expression pattern analysis were then con-ducted.The results showed that the CsPHY proteins had 1 005 to 1 345 amino acids with the molecular weights as 111.13 to 148.78 kDa,the theoretical isoelectric points as 5.79 to 6.70,and the phosphorylation sites as 68~113.CsPHYA2 and CsPHYB2 were localized in chloroplasts,and CsPHYA1 and CsPHYB1 were in mito-chondria,while CsPHYE might localized in cytoplasm.All the five CsPHY sequences contained PAS domain,GAF domain,and phytochrome region.Furthermore,CsPHY was closely related to the Actinidia chinensis Ad-PHYB.In addition,light quality could influence the expression pattern of CsPHY genes,and red light signifi-cantly upregulated the expression level of CsPHYA1 and CsPHYB2.Based on previous research results,it was inferred that the upregulation of CsPHYA1 and CsPHYB2 under red light was the main reason for greening of Huangjinya leaves.关键词
