摘要

Abstract

To construct a full-length transcriptome database of Atractylodes chinensis(DC.)Koidz and mine molecular markers,transcriptome sequencing was performed using single molecule real-time(SMRT)technology with roots,stems and leaves as experimental materials.The key enzyme genes potentially involved in terpenoid biosynthesis were identified,and the applicability of SSR molecular markers in A.chinensis and A.lancea was preliminarily evaluated.The results showed that after de-redundancy analysis,a total of 14 556 high-quality full-length transcripts were obtained,of which,13 962(95.92%)were successfully annotated in five public databases(KEGG,GO,Nr,Uniprot and Pfam).KEGG annotation analysis indicated that 11 907 transcripts were assigned to 126 pathways with the largest number of transcripts annotated to metabolism-relat-ed pathways as 6 606.A total of 133 transcripts involved in terpenoid and polyketide metabolisms were identi-fied,and 12 transcripts were mapped to the sesquiterpenoid and triterpenoid biosynthesis pathways;59 lncR-NAs and 14 438 CDS sequences were obtained,4 443 SSR loci and 688 transcription factors were detected,and the GRAS transcription factor family owned the most members.Some randomly selected SSR molecular markers were successfully amplified in A.chinensis and A.lancea.These findings could provide new insights into the molecular mechanisms underlying sesquiterpene biosynthesis in Atractylodes,as well as provide bases for molecular marker development and mining and utilization of functional genes.
