补肾活血方对子宫内膜炎大鼠的保护及其对JAK2/STAT3信号通路调控的实验研究
Experimental Study on the Protection of Bushen Huoxue Decoction on Rats with Endometritis and Its Regulation of JAK2/STAT3 Signaling Pathway

Abstract
Objective Based on the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway,the protective mechanism of Bushen Huoxue formula(BHF)against the lipopolysaccharide(LPS)-induced endometritis in a rat model was thoroughly investigated.Methods 60 SD rats were randomly divided into 6 groups,10 rats in each group:Sham operation group,model group,positive control group(given dexamethasone acetate,dose of 2.5 mg/kg),and low dose group(L-BHF),middle dose group(M-BHF)and high dose(H-BHF)group,and the doses of 7.8,15.6,31.2 g/kg).HE staining was used to observe the pathological changes of endometrium;MPO activity was determined by colorimetry;The levels of inflammatory cytokines IL-6,IL-1β,TNF-α,IL-4,and IFN-γ were measured using the enzyme-linked immunosorbent assay(ELISA);Moreover,flow cytometry was employed to detect the Th1 and Th2 cells within the peripheral blood samples,JAK2/STAT3 signaling pathway related proteins were detected by western blot.Results Compared with the sham operation group,the number of inflammatory cell infiltration,MPO activity,TNF-α,IL-1β and IL-6 levels,Th1 and Th1/Th2 levels,p-JAK2/JAK2 and p-STAT3/STAT3 expression in the model group were significantly increased,and Th2 was significantly decreased(P<0.05).Compared with the model group,the number of inflammatory cell infiltration,MPO activity,TNF-α,IL-1β and IL-6 levels,Th1 and Th1/Th2 levels,p-JAK2/JAK2 and p-STAT3/STAT3 expression in the L-BHF group,M-BHF group and H-BHF group were significantly decreased,and Th2 level was significantly increased(P<0.05).Conclusion Bushen Huoxue formula can improve endometrial inflammatory symptoms by blocking JAK2/STAT3 signal transduction,inhibiting the release of pro-inflammatory factors,and regulating Th1/Th2 immune balance.关键词
