Functional analysis of growth promotion and disease control in Streptomyces alfalfae 5406 based on multi-omics

Abstract
Streptomyces alfalfae 5406,isolated from alfalfa rhizosphere soil,is a biocontrol strain with dual functions of plant growth promotion and disease control.This study aims to investigate the impact of tomato seedlings on the metabolites and gene expression of S.alfalfae 5406,identify growth-promoting and disease-resistant metabolites along with their regulatory genes,and assess the metabolite antibacterial activity against Ralstonia solanacearum.A co-culture system involving S.alfalfae 5406 and tomato seedlings was established.Integrated metabolomic and transcriptomic analyses were conducted to identify key differential metabolites and expressed genes.The functions of bioactive metabolites were validated through in vitro antibacterial assays.The results indicate that S.alfalfae 5406 is capable of synthesizing indole-3-acetic acid(IAA)through the tryptophan-dependent indole-3-acetamide(IAM)pathway.Additionally,it can produce gallic acid,methyl gallate,and daphnetin,all of which exhibited inhibitory activity against R.solanacearum.S.alfalfae 5406 demonstrates dual capabilities of promoting plant growth and suppressing diseases.It enhances plant growth and combats pathogens through various mechanisms,including IAA synthesis and bacteriostatic compound production.This research lays a groundwork for the potential development of agricultural biocontrol agents utilizing S.alfalfae 5406.关键词
