Effects of Induced Molting on Reproductive Function and Hypothalamic Transcriptome Changes in Aging Cocks

Abstract
This study aimed to evaluate changes in reproductive-related phenotypes and hypothalamic transcriptomic features in aged breeder roosters following fasting-induced molting(FM)and to analyze the association between them.A total of 30"Yufen No.1"H-line roosters,aged 60 weeks,were randomly assigned to two groups according to the experimental design:a control group consisting of 10 birds and a treatment group consisting of 20 birds.The body weight,serum levels of high-density lipoprotein cholesterol(HDL-Cho)and low-density lipoprotein cholesterol(LDL-Cho)were recorded for the treatment group on the day before fasting(F0),on the 15th day of fasting(F15),and on the 39th day of refeeding(R39).Additionally,hypothalamic tissue was collected for transcriptome sequencing(RNA-seq).On R39,semen volume,sperm density,sperm motility,and serum hormone levels were evaluated for all roosters in each group.The results demonstrated that FM significantly enhanced sperm motility(P<0.05)and density(P<0.05)in the roosters.Although there was an increasing trend in the levels of follicle-stimulating hormone(FSH),luteinizing hormone(LH),and testosterone(T),the differences were not statistically significant(P>0.05).Following refeeding,body weight returned to normal,and LDL-Cho levels showed a significant increase(P<0.05).RNA sequencing identified a total of 14 752 genes,of which 7 030 exhibited significant differential expression.These genes were primarily involved in pathways such as neuroactive ligand-receptor interaction,ECM-receptor interaction,cell adhesion molecules,and adherens signaling.During fasting,neuropeptides such as AGRP,NPY,POMC,and GAL were upregulated,whereas,upon refeeding,neurodevelopment-related genes like GPM6B and BASP1 were upregulated.Immune-related genes such as CXCL12 and GRP were downregulated during fasting.Protein interaction analysis revealed potential hub genes,including AGT,NTS,TRHR,and CCK.The findings of this study suggest that the hypothalamus may play an indirect role in the restoration of reproductive function during the FM process,potentially mediated by transcriptional alterations in neuropeptides and metabolic signaling pathways.This research provides preliminary molecular insights into the reproductive recovery mechanisms induced by molt in aging cocks.关键词
