JunB regulates brown adipocyte differentiation and thermogenesis in mouse C3H10T1/2 cells

Abstract
Objective We investigated the effect of silencing JunB on the differentiation of C3H10T1/2 cells into brown adipocytes and thermogenic capacity.Methods C3H10T1/2 cells were cultured in vitro and transiently transfected with small interfering RNA(siRNA)to establish a JunB silencing model.The best JunB-siRNA group was selected by Cell Counting Kit-8(CCK-8)and Western blot analysis.Differentiation was induced by a cocktail method,and blank control(Control),induced differentiation model(DM),and induced differentiation model with JunB silencing groups(DM+JunB-siRNA)were created.Lipid droplet formation was analyzed by Oil Red O staining.Western blot analysis and immunofluorescence staining were used to detect uncoupling protein 1(UCP1)and peroxisome proliferator-activated receptor γ coactivator-1α(PGC-1α)protein expression,and a JC-1 kit was used to detect mitochondrial membrane potential.Results JunB-siRNA had no significant effect on cell viability(P>0.05),with JunB-siRNA Ⅰ silencing having the best effect(P<0.001).JunB silencing increased lipid droplet accumulation in brown adipocytes.UCP1 and PGC-1α expression was up-regulated(P<0.05,P<0.001)and mitochondrial membrane potential was enhanced.Conclusions JunB silencing increased the lipid droplet content and thermogenesis of brown adipocytes,likely through improving mitochondrial function.关键词
