堇叶碎米荠高效离体再生体系构建与优化
Establishment and Optimization of an Efficient in Vitro Regeneration System for Cardamine circaeoides

Abstract
To establish an efficient in vitro regeneration system for Cardamine circaeoides,the seeds of Cardamine circaeoides were used as the material for obtaining sterile tissue culture seedlings.Based on the MS medium,by adjusting the types and concentrations of plant growth regulators,the effects of differences in explant sites and hormone ratios on the formation of callus,the differentiation of ad-ventitious buds,and the induction of rooting were studied.The results showed that the best disinfec-tion method for the seeds was 0.1%HgCl2 for 7 minutes;the best explant for inducing callus was the hypocotyl,and the best induction medium was MS+6-BA0.5 mg/L+NAA0.1 mg/L,with an induc-tion rate of 75.93%.The best medium formula for the differentiation of adventitious buds was MS+6-BA 1.0 mg/L+NAA0.5 mg/L,with a differentiation rate of 85.93%.The rooting medium was 1/2 MS+IBA 1.0 mg/L,with a rooting rate of 98.52%.After the tissue culture seedlings were transplan-ted to the substrate,they grew fast and robustly,and the transplant survival rate reached 85%.关键词
