双参芍胶囊通过抑制TGF-β/Smad和NF-κB通路的激活改善模型大鼠肝纤维化病变的实验研究
Effect of Shuangshenshao Capsules on Ameliorating Rat Liver Fibrosis by Inhibiting Activation of TGF-β/SMAD and NF-κB Pathways

Abstract
Objective:To explore the therapeutic effect and mechanism of Shuangshenshao Capsules(双参芍胶囊)on liver fibrosis.Methods:Sixty-eight Sprague Dawley(SD)rats,half male and half female,were randomly divided into a normal control group(n=14)and a model group(n=54).The model group was subcutaneously injected with 40%carbon tetrachloride(CCL4)for 10 weeks to construct an animal model of liver fibrosis.The successful model rats were randomly divided into a normal control group,a model control group,Shuangshenshao Capsules groups of 1.1 g/kg,0.6 g/kg,and 0.3 g/kg,and a Compound Glycyrrhizin Tablets group of 24 mg/kg.Rats were orally administrated with corresponding drugs or pure water.The level of alanine aminotransferase(ALT),aspartate aminotransferase(AST),hy-aluronic acid(HA),laminin(LN),type III procollagen(PCIII),type IV collagen(ColIV),interleukin(IL)-1β,and IL-6 in serum was detected by biochemical kits.Hematoxylin-eosin(HE)staining and Masson staining were used to ob-serve the pathological changes in liver fibrosis in rats.The expression level of transforming growth factor-β1(TGF-β1),mothers against decapentaplegic homolog 3(SMAD3),alpha-smooth muscle actin(α-SMA),SMAD7,P65,and inhibitor of nuclear factor kappa-B alpha(IκB α)in the liver tissue of each group was detected by Western Blot.Results:Com-pared with those in the normal control group,the level of AST,ALT,IL-1β,IL-6,HA,LN,PCIII,and Col IV in the model control group was significantly increased(P<0.01).The protein expression of TGF-β1,SMAD3,α-SMA,P65,and IκB α was upregulated(P<0.05 or P<0.01),while the protein expression of SMAD7 was downregulated(P<0.01).Com-pared with that in the model control group,the content of AST and ALT in the Shuangshenshao Capsules groups of 1.1,0.6,and 0.3 g/kg and the Compound Glycyrrhizin Tablets group of 24 mg/kg was significantly decreased(P<0.01 or P<0.05).The content of LN,IL-6,IL-1β,and HA was significantly decreased(P<0.05 or P<0.01),and the protein expression of TGF-β1,SMAD3,α-SMA,P65,and IκB α was downregulated(P<0.05 or P<0.01).In liver tissue,mild scattered inflammatory cell infiltration and a narrowed distance between collagen fibers in the hepatic lobules were ob-served,and a small amount of fat degeneration was found in hepatic cells.The cell apoptosis rate at the early stage was significantly increased in the Shuangshenshao Capsules groups of all doses(P<0.01).Conclusion:Shuangshenshao Capsules can improve liver fibrosis in model rats.The mechanism may be related to inhibiting the activation of TGF-β/SMAD and nuclear factor kappa B(NF-κB)pathways and reducing the inflammatory response of liver.关键词
